Some organisms (members of <em>Cloistridium<\/em>, <em>Desulfotomaculum<\/em>, and <em>Bacillus<\/em>) may exist as vegetative cells or spores in order to survive a harsh environment.<\/p>\n<p><strong>Sporogenesis<\/strong> allows an intracellular <strong>endospore<\/strong> to develop which is surrounded by layers of <strong>spore coats<\/strong> (very tough layers to protect endospore). As the external conditions worsen, the vegetative cell may release the endospore which allows the endospore to become an independent <strong>free spore<\/strong>. When external conditions become more favorable, the free spore may revert back (germination) to a vegetative cell.<\/p>\n<p>Spore staining uses 2 reagents<\/p>\n<p>The <strong>primary stain<\/strong> is malachite green (with heat). This will color the spores.<\/p>\n<p>The <strong>decolorizing agent<\/strong> is water. This will decolorize the vegetative cells.<\/p>\n<p>The <strong>counterstain<\/strong> is Safranin which will colorize vegetative cells red.<\/p>\n<p><strong>Clinical significance<\/strong>. Helps to identify members of\u00a0<em>Cloistridium<\/em>, <em>Desulfotomaculum<\/em>, and <em>Bacillus <\/em>such as <em>Bacillus anthracis<\/em> (anthrax) and <em>Cloistridia<\/em> bacteria (tetanus, gas gangrene, food poisoning and pseudomembranous colitis).<\/p>\n<p><b>Summary of procedure.<\/b><\/p>\n<ol>\n<li>Prepare a smear and heatfix.<\/li>\n<li>Flood slide with malachite green and place over a beaker of warm water (on a hotplate).<\/li>\n<li>Steam for 2-3 minutes.<\/li>\n<li>Cool slide then gently flush with deionized water.<\/li>\n<li>Flood slide with Safranin for 30 seconds.<\/li>\n<li>Gently flush slide with deionized water.<\/li>\n<li>Blot dry with bibulous paper.<\/li>\n<\/ol>\n<p>Reference<\/p>\n<p>Cappuccino,\u00a0J.\u00a0G., &amp; Welsh,\u00a0C.
